Getting the Lock spell, also known as Alohomora, in Hogwarts Legacy is actually part of the story! You'll need to progress through the main storyline until you reach a mission called 'The Caretaker's Lunar Lament'. This quest is given by Gladwin Moon, the Hogwarts caretaker. Complete his tasks, and you'll learn Alohomora as a reward, letting you unlock those pesky locked doors around Hogwarts.

Alohomora isn't a spell you can just pick up whenever. Stick to the main quests until 'The Caretaker's Lunar Lament' pops up. Trust, it's worth the wait for all the secret stuff you can unlock later.
