Easiest way? Play 800 Vs. matches. After that, Luigi will challenge you and you just have to win. Simple and effective!

It worked for me! Complete Adventure Mode ending a stage with a '2' in the timer. Luigi pops up, you beat him, and voilà! Luigi's yours.
