To get mastery in Blox Fruits, you just need to keep using your fruit's abilities or your sword to defeat enemies. The more you use them, the higher your mastery level will be!
In my experience, fighting bosses and farming NPCs was the fastest way to rack up mastery. Patience and persistence pay off!
