Honestly, just grind those new weapons. Crafting and leveling up a ton of gear is the quickest way to rack up mastery points.
Do Sanctuary Onslaught for fast Affinity gains. It's a bit repetitive, but efficient. Trust me, I maxed out like 10 weapons there in no time.
Focus on leveling up new weapons, Warframes, and companions. Each one gives you mastery points the first time you rank it up! Also, don't forget to hit those daily quests and alerts for some extra mastery juice.
