You gotta be on the Pacifist Route. After you defeat Asgore, Alphys will call you. As you backtrack, you'll find the entrance to the True Lab in her lab.

First, make sure you haven't killed anyone (seriously, not even Jerry). Then, after beating Asgore and Flowey, backtrack to Alphys's lab to trigger the True Lab event.

Complete the Neutral Route first, then do another playthrough on the Pacifist path. Once you reach the end, you'll get a phone call from Alphys instructing you to return to her lab. That's where you'll find the True Lab entrance.

Just play the Pacifist route, and go through Alphys' lab after you beat the king. Simple as that.
