Just make sure you're on the True Pacifist Route! Beat Asgore, spare Flowey, then Alphys will call you. Follow the path in the elevator to find the lab!

To get to the Underground Lab in Undertale, you need to complete the True Pacifist Route. After sparing Asgore and Flowey in the Neutral Route, Alphys will call you with an invitation. Head back to the elevator that leads to New Home, and then take the new path to the lab!

Honestly, just focus on not killing anyone and being friends with everyone, especially Alphys. After the Neutral Ending, Alphys will call and open the way.
