Put a boss like Bowser in a room with some obstacles (like blocks, firebars, or Thwomps). Add a couple of power-ups to help the player out. Voila, you're done!

Start by choosing a main boss, like Bowser or Bowser Jr., then build an arena around them with plenty of hazards and power-ups to create a tough but fun fight. Don't forget to use checkpoints!
