To make vertical levels in Super Mario Maker 2, open Course Maker, start a new course, and select the vertical sub-area from the options. Adjust the height using the scroll bar on the right!

You just need to place your starting point on the bottom, then scroll the screen upwards to build vertically. Easy peasy!
