You can't directly play Fallout Shelter on Amazon Fire TV 'cause it's not supported. BUT, there's a workaround: if you've got a compatible smartphone or tablet, just download the game there and use a screen mirroring app to cast it to your Fire TV. It's not the smoothest way to play, but hey, it works if you're really itching to see your vault on the big screen.
Sadly, Fallout Shelter isn't available on Amazon Fire TV. Would've been awesome though!
Tip from a long-time player here: after getting Fallout Shelter up on your Fire TV, consider using a game controller if you have one. It makes navigating through the vault and managing your dwellers so much easier than the Fire TV remote!
