The Xbox 360 doesn't support playing games directly from a USB. Your best bet is to copy the game content onto your hard drive. Use the USB for game saves, profiles, and DLC instead.
Unfortunately, it’s a no-go on playing directly from USB!
What I usually do is use an external hard drive for storage and then transfer the game to the internal hard drive of the Xbox 360. This method works well and keeps everything smooth. Just remember to format your hard drive first in the console settings!
