How to Set Trade Routes in Fallout 4
Setting trade routes in Fallout 4 is pretty straightforward but super essential for your settlements to thrive. First, you gotta have the Local Leader perk (which requires you to be at least level 14 and have a Charisma level of 6). Once you've got that, head over to one of your settlements, enter Workshop mode, and find a settler you wanna assign. Press the supply line option (it's Q on PC, R1 on PS4, and RB on Xbox One), then just pick the settlement you wanna create a route with. Boom, you're done! Trade route is set, and your settlements are now sharing resources.

Actually setting trade routes is a waste of time. Just stuff everything into one settlement and call it a day. Who wants to micromanage a post-apocalyptic wasteland anyway?
From personal experience, managing trade routes well can seriously up your settlement game. Make sure to evenly distribute your settlers though, so every settlement can grow. It's also a good idea to connect settlements in close proximity to each other to make a network. Saves you a lot of hassle in the long run!
