First thing, ya gotta have the Local Leader perk (it's in the Charisma tree and you need at least Charisma level 6 to unlock it). Once you've got that, head over to one of your settlements. Find a settler you wanna assign to the supply line, then enter the Workshop mode. Look for the 'Supply Line' option when you're highlighting the settler (it'll pop up at the bottom of your screen, the button depends on your platform). Press it, then pick the settlement you wanna link up. Boom, you're in business.

Supply lines? Overrated. Just hoard everything in one place and fast travel. Much simpler.

Just get the Local Leader perk, then in Workshop mode, click on a settler and send them to another settlement as a supply runner. Easy.
