To set up trading routes between your settlements in Fallout 4, you first need to unlock the Local Leader perk, which becomes available when you have a Charisma level of 6. Once you have it, go to your settlement's workshop view, highlight a settler, and you'll see an option to assign them as a supply line to another settlement. This way, resources will be shared between the connected settlements!

Just get the Local Leader perk and start assigning settlers to trade routes. Easy peasy lemon squeezy. They'll handle the rest.

Actually, you don't directly trade items between settlements. The supply lines just share resources for building and crafting. If you want specific items in another location, you'll have to carry them yourself.
