I unlocked characters by playing all the single-player story modes and doing online matches to gather Fight Money. Took a while, but it felt rewarding!

Grind that Fight Money! Complete Daily Missions, Trials, and Story Modes to rack up enough to buy new characters. Or, if you're impatient, just throw money at the screen and purchase them from the store.
