Just collect 11 gold medals in the Blue Sphere bonus stages and there you go, & Knuckles mode unlocked.

To unlock & Knuckles mode in Sonic Mania, just enter the secrets menu by collecting enough gold medals in Blue Sphere stages. Then select your game file, hit the secrets menu, and toggle & Knuckles on. Voila, your new echidna buddy will be tagging along!
