You can unlock Lucario in Super Smash Bros Brawl by playing through the Subspace Emissary mode until you encounter and defeat him. Alternatively, you can unlock him by playing 100 Brawl matches or completing all 5 Target Smash levels with any character!

To unlock Lucario, just play through 100 Brawl matches or beat him in the story mode. Easy peasy!
I unlocked him by just playing a ton of Brawl matches with my friends, but the Subspace Emissary route works too!
Honestly, just go through the Subspace Emissary. It's more fun and you unlock other characters along the way.
