Honestly, the fastest way is playing through Subspace Emissary. You'll unlock almost everyone by the end of it.
Just grind out vs. matches if you're not into the Subspace Emissary. After a certain number, you'll get challenged by different characters!
Use a guide! Some characters have really specific requirements, like playing a bunch of Brawl matches or completing certain challenges.
