First, make sure your Xbox 360 is connected to the internet. Then, launch Lego Dimensions while connected, and the game will automatically check for and prompt you to download any available updates.

I had to ensure my Xbox Live account was active and the Xbox 360 connected to my home network. Then, launching Lego Dimensions took care of the rest as it downloaded the update on its own.
