As of my last check, Rockstar hasn't spilled all the beans on GTA 6 yet, but multiplayer has always been a big part of the GTA series. GTA 5 and GTA Online set the bar pretty high, so it's a safe bet that GTA 6 will have some form of multiplayer or 2 player mode. Keep an eye out for official announcements though!

Nope, GTA 6 being 2 player isn't confirmed. Rockstar games are tight-lipped as always. Would be cool though!

Honestly, imagine GTA 6 without a 2 player option. That'd be like peanut butter without jelly. Given Rockstar's history with GTA Online, they'd be missing a huge trick not to include it.
