So, if you've got a ton of wood in Sanctuary but need it in The Castle, just set up a supply line, and voila! You can build that fancy schmancy defense turret without having to lug all that wood across the Commonwealth. Basically, it makes managing your resources way easier and helps you become the wasteland tycoon you always wanted to be.

Supply lines? Oh, those are the magic bridges that make your hoarding problem look like strategic resource allocation. You assign a settler to connect two settlements, and suddenly they're sharing crafting materials like it's a post-apocalyptic communal living situation. You haven't lived until you've seen your adhesive empire stretch across the map.
They're essentially the game's way of telling you that you don't have to carry every tin can and screwdriver you find back to your main base by yourself. Set up a supply line, and your settlers will do the heavy lifting, figuratively speaking. Makes building and crafting a heck of a lot more convenient.
Honestly, never used them. Just hoarded everything at Red Rocket. Worked out fine for me.
