I'd say the Moon Lord, but don't sleep on the Empress of Light during the day. It's a whole different ball game when you fight her then, with everything one-shotting you. Not for the faint of heart!
Honestly, Duke Fishron can be a real pain if you're not properly prepared. Fast, hits hard, and doesn't give you much room to breathe. Not the ultimate hardest, but definitely deserves an honorable mention.
Moon Lord, no contest. You think you're all set with your armor and weapons, then BOOM, reality check. Definitely a challenge that tests all your Terraria skills.
