Vaults open as soon as you use the keycards you find in the game. They're usually with bosses or in rare chests.

Vaults in Fortnite only open when you have a keycard, which you can get from defeating certain bosses or enemies, usually found at key strongholds on the map. Good luck hunting!

Just get a keycard and the vaults will open. No keycard, no vault.
