Just head straight to Fort Defiance if you want to start the Brotherhood of Steel questline. Can't miss it, it's that creepy looking building in the Cranberry Bog.
If you're looking to join the Brotherhood of Steel, your first step should be to listen for Brotherhood emergency broadcasts on your Pip-Boy. This will eventually lead you to start the 'Defiance Has Fallen' quest at Fort Defiance. P.S.: Prepare for some intense action and deep lore!
