Actually, synths aren't a thing in Fallout Shelter. You're mixing up the wider Fallout lore. In Fallout Shelter, you're managing a vault and dealing with raiders, resources, and dwellers. The whole business with the synths and the Institute is part of the mainline Fallout games, especially Fallout 4.

The synths in the Fallout universe, including Fallout Shelter, were created by the Institute. The Institute is a secretive organization with advanced technology, based underground in what remains of the Massachusetts Institute of Technology (MIT). They're pretty much the boogeymen of the Commonwealth, creating synths (synthetic humans) that can sometimes pass for real humans!
