Tsunade is hands down the strongest kunoichi in Naruto Shippuden! Not only is she a legendary Sannin, but her monstrous strength and healing abilities are unmatched. Plus, she can wreck buildings like it's nobody's business!

Mei Terumi, the Mizukage, deserves a mention! Her Lava and Boil Release techniques are OP and she can hold her own against Akatsuki members. Plus, she runs an entire village – strong and badass!
