Disable any overlays you might have running. Steam overlay, for instance, can sometimes cause conflicts. Turn it off and see if that helps!

Try verifying the game files through Steam. Sometimes a corrupted file can mess things up. Go to your library, right-click on The Binding of Isaac, select Properties, then Local Files, and click 'Verify Integrity of Game Files'.
