Tsunade's absence in Boruto is likely due to her retirement from active duty. Her time as Hokage and a legendary ninja ended with the close of Naruto Shippuden.
From what I gather, Tsunade isn't in Boruto because it's time for new characters to shine. She's had her legendary run!
Because she's retired and letting the new generation take over. Simple as that.
