Check your Xbox Live account status! Sometimes if your subscription lapses, you can't upload clips until it's renewed. Happened to me once, drove me nuts!

Make sure your privacy settings aren’t blocking uploads. Go to Settings > Account > Privacy & Online Safety, and check your settings.
