You can't directly play with 100 players in Among Us, but you could set up multiple games simultaneously and rotate players. Just think of it as a mega tournament!

Sorry to burst your bubble, but Among Us officially supports a max of 15 players per game. Throwing 100 players in there would probably break the spaceship!
