To get the bus running in Stardew Valley, you gotta unlock the Calico Desert. It's simpler than it sounds: head to the Community Center and complete the Vault Bundle. This involves donating a bunch of gold to the vault. The amounts will total 42,500G. Once you pay up, the bus to Calico Desert starts running the next day.
