To unlock the bus in Stardew Valley, you gotta complete the Vault bundle at the Community Center. It means you have to donate a bunch of gold to fix up the room. Prices range from 2,500G to 25,000G for each bundle. Once you've done them all, the bus to Calico Desert gets up and running again.
